It had been allocated a place on the final Saturday of the event in recent years, but will revert to a day earlier next year.
The final Friday of race week is a bank holiday in the Isle of Man.
The Senior race was cancelled in 2025 because of inclement weather and red-flagged on the second lap this year, after being moved forward a day in the schedule.
Attempts to run the race the following day proved unsuccessful in light of bad weather, and the result was taken from the positions at the end of lap one of Friday's race, with Dean Harrison being declared the winner.
Other changes to the 2027 race programme include the Superbike race moving from Sunday to Saturday on the opening race weekend and just one Superstock race being staged, rather than the two which were scheduled for recent years.
A total of nine races will form the schedule, rather than the 10 which were pencilled in for recent editions of the meeting.
The first Supersport event is set for Sunday 6 June, one day after the six-lap Superbike opener.
Monday 7 June is listed as a contingency day, with racing resuming on Tuesday 8 June with the solitary Superstock race and sidecar race one.
The second Supersport race and the first Sportbike outing are slated for Wednesday 9 June, with Thursday a contingency day.
The 100th edition of the Senior TT is scheduled to take place on Friday 11 June, with Saturday providing a contingency opportunity should the race be affected by weather or other delays.
Race week will conclude on Saturday, 12 June, with the second sidecar and second Sportbike races.
The Superstock and Sportbike classes will be staged over four laps next year, instead of three, with sidecars reverting from three to two laps of the Mountain Course.
Qualifying will take place from Monday, 31 May to Friday, 4 June and will retain the same format and session structure used in 2026.
